Narrative
Originally M.truncatula var. longeaculeata. R108 is a generic name for the accession whose origin remains uncertain, it was probably collected in the Middle East. This ecotype has been used by several laboratories (In France, mainly by INRA Toulouse and ISV-CNRS GIF sur Yvette) in the 1990s because of its ability for in vitro regeneration. To my knowledge, the original accession (collected in the wild) was never maintained. The available samples came all from multiplications in laboratories, often after several cycles of in vitro culture. The seeds you received, R108-C3, came from one multiplication in glasshouse at INRA Montpellier of one lot received from ISV-CNRS in 1998. This line was regenerated after three cycles of in vitro culture, hence the post-fix C3 following R108. It is difficult to answer if R108-C3 is identical to R108-1 (C3), but I suppose yes. Our line (R108-C3) is identical to those used by the ISV-CNRS, which is one of the main users of this line. ISV-CNRS (Pascal Ratet) has also produced several collections of lines transformed by Agrobacterium from this line R108-C3 that represents the non-transformed control.
